SS-31 (Elamipretide / MTP-131 / Bendavia) is an aromatic-cationic tetrapeptide (D-Arg-DMT-Lys-Phe-NH2) developed by Szeto and Schiller. The mechanistic literature documents that it crosses the outer mitochondrial membrane and binds selectively to cardiolipin on the inner membrane (Szeto HH, Br J Pharmacol 2014, PMID 23647488), with stabilization of respiratory chain supercomplexes (I, III, IV), preservation of ATP synthase, reduction of electron leakage, and decreased ROS without collapse of the ΔΨm potential in experimental models.
