What is it and how does it work?

Tesamorelin is a stabilized synthetic analog of human growth hormone-releasing hormone (GHRH). It binds to the GHRH-R receptor on pituitary somatotrophs and stimulates pulsatile release of endogenous growth hormone, with documented increases in serum IGF-1 in clinical studies. The literature describes a preferential effect on abdominal visceral adipose tissue in cohorts with HIV-associated lipodystrophy.

Clinical evidence: Falutz et al. (NEJM 2007, PMID 17984164) reported a reduction in visceral fat in patients with HIV-associated lipodystrophy following treatment with 2 mg/day subcutaneous (SubQ) 15-20% for 26 weeks. Stanley et al. (JCI 2014, PMID 24911148) reported improvement in hepatic steatosis in nonalcoholic fatty liver disease. Research use only—no FDA-approved human use outside the specific indication of HIV-associated lipodystrophy (Egrifta).
